Glutaredoxins and thioredoxin peroxidase involved in defense of emamectin benzoate induced oxidative stress in Grapholita molesta

Glutaredoxins (Grxs) and thioredoxin peroxidases (Tpxs) are major antioxidant enzyme families involved in regulating cellular redox homeostasis defense of enhanced oxidative stress through scavenging reactive oxygen species (ROS). However, the functions these enzymes have not been reported oriental fruit moth, Grapholita molesta (Busck), a worldwide pest stone pome fruits. Here, we identified four new genes, GmGrx, GmGrx3, GmGrx5, GmTpx which were induced by exposure with emamectin benzoate, commonly used biopesticide for G. control. Other environmental factors (low high temperatures, Escherichia coli Metarhizium anisopliae) also significantly expression genes. After GmGrx or silenced RNA interference (RNAi), percentage larval survival to benzoate decreased, demonstrating that protecting from stresses benzoate. Furthermore, enzymatic activities superoxide dismutase (SOD) (except GmTpx) peroxidase (POD), as well contents hydrogen peroxide metabolites ascorbate. Taken together, our results suggest may play critical roles defense. Specially, contribute damage excessive ROS molesta. Our findings provided theoretical basis understanding insect glutaredoxin systems.
